The Dysregulation of Cyclin Dependent Kinase Regulators Role in SV40 Related Renal Cell Carcinoma

Abstract

Aim: This research using P.C.R. and immunohistochemical techniques to assess the assistance of SV40 polyomavirus infection to the development of renal cell carcinoma in patients from the province of Al-Najaf. Method: This study included (75) (meaning: 45 males and 30 females, ages ranging from 22 to 70 years) paraffin-impeding block tissues of renal cell carcinoma from AL-Sader Medical City archives and few archives of private histopathology laboratories in this governorate, was planned as a cross-sectional study to discovered SV40 in renal cell carcinoma. Bu utilizing polymers chain reaction (P.C.R.) to detected both nucleic acid of SV40 and immunohistochemistry (IHC) by expression state of Cyclin Dependent Kinase Regulators (KAP or cyclin-dependent kinase inhibitor 3 (CDKN3) & Cyclin E1 markers), the data cover the period from January 2016 to December of the similar year. RCC block usually was identified by hematoxylin and eosin stains. Results: The Immunohistochemistry technique (IHC) results showed a higher positive percentage for the KAP or CDKN3 marker and a lower positive percentage for the Cyclin E1 marker. Furthermore, it was discovered that the percentage of clear cells was higher at 42 (56%), grade I was higher at 31 (41.3%), and tumor stage type I was higher at 25. PCR methods yielded positive results in 20 (26.7 percent of 75 cases) of block tissues in the RCC patient. Conclusion: The results of these study showed that Simian Virus 40, in especial its Large T Antigen (T-ag), that affected CDK regulators and disrupts the delicate equilibrium of cell cycle control. Consequently, this study recommends a possible association between SV40 polyomavirus and the progress of renal cell carcinoma. This study recommends by using routine I.H.C. and P.C.R. techniques for detection of RCC.
